It's Colt's job to take out eight Visionaries throughout the island before midnight, since leaving even one alive would reset the time loop and destroy all of his hard work, so he's got to do it quickly. Colt will be reset to the beginning of the cycle if he dies before taking out all eight targets. Stealth, parkour, assault skills, firearms, gadgets, and abilities are all at the disposal of the player in Arkane's earlier Dishonored and Prey games. Colt's targets must be avoided or taken out, and they must decide out the best sequence in which to use weapons, physical attacks, or other environmental techniques to remove them.

Players in Deathloop have a lot of leeway when it comes to taking down these eight targets in one loop since the timer isn't set in stone. Moving between the island's four districts (Updaam, Karl's Bay, Fristad Rock, and The Complex) causes time to move forward throughout the four times of the day (Morning, Noon, Afternoon, and Evening). The player's activities in one district might alter the routines in other districts, depending on the time of day the player visits the area. When playing as Julianna, an agent entrusted with preserving the time loop and eliminating Colt, you may switch roles and play as yourself. Taking on this role allows the player to infiltrate another's game and perhaps disrupt their gameplay. When Colt Vahn awakens from a dream in which he is killed by an unknown lady, he has no idea who or where he is or how he got there. However, he gets messages and meetings from various versions of himself, who tell him to break the time loop he is in. By killing all eight Visionaries before time loops, he may accomplish this. Additionally, Julianna Blake, the chief of security, notifies the Visionaries (and their followers) of Colt's plot and demands for him to be tracked down, complicating things even more. Julianna also tries to break the loop by taunting Colt, despite her efforts to keep him from doing so. At some point Colt discovers that his memories are preserved throughout loops, giving him an advantage in preparing for his eventual aim of breaking the loop: he can better plan and strategize. Moreover, he discovers that Julianna's memories seem to be preserved throughout loops.

Despite the fact that Colt has devised a plot to murder seven of Blackreef's Visionaries, Julianna remains the most elusive by hiding in the Loop, the structure that controls the time loops. It is only possible to get to the Loop using a rocket jet that was left behind by the military, so he starts searching among the island's several abandoned bunkers. One of the participants of the initial military mission to Blackreef known as Operation Horizon, he ultimately learns that he was unintentionally transported into the future due to an experiment that went wrong. Lila's ex-boyfriend Colt then joined the AEON Program in the hopes of reuniting with her in the past. While in the future, he realizes that Julianna is in reality his own daughter. After activating the rocket aircraft, Colt reaches the Loop and faces Julianna. In Julianna's version of events, things began to go awry when Colt began killing her in every loop in an attempt to liberate her from the AEON Program. Her hatred towards Colt steadily intensified, ending in her hunting him down in every loop. If you murder her and break the loop, you'll have to face an uncertain future. If you spare her, you'll be able to live endlessly via the loops.
